Expression of MARK2 in hepatocellular carcinoma and its effect on the migration of hepatocellular carcinoma cells

摘要: 背景与目的:肝细胞癌是我国常见恶性肿瘤之一,其发生机制目前尚未明确。该研究旨在探讨微管亲和性调节激酶2(microtubule affinity-regulating kinase 2,MARK2)在肝癌中的表达及其对肝癌细胞迁移能力的影响。方法:通过组织芯片和免疫组织化学法对肝癌组织中MARK2的表达进行分析;将MARK2基因真核表达载体及其空载体转染至肝癌HepG2细胞中,采用蛋白质印迹法(Western blot)检测转染后MARK2蛋白表达水平;采用Transwell实验及细胞划痕实验检测MARK2对HepG2细胞迁移能力的影响;通过癌症基因组图谱(the Cancer Genome Atlas,TCGA)分析MARK2在肝癌组织和正常组织中的表达差异,及其与肝癌患者临床病理特征的关系;运用网络在线生存期分析软件Kaplan-Meier plotter分析MARK2表达与肝癌患者生存预后的关系。结果:MARK2在肝癌组织中表达明显高于癌旁组织,肝癌组织中MARK2高表达与TNM分期及病理分级显著相关(P<0.001);生存分析结果表明,MARK2高表达患者5年生存率明显低于MARK2低表达组(P<0.01);将MARK2基因真核表达载体转染肝癌HepG2细胞后,MARK2蛋白表达上调,肝癌细胞的迁移能力增强。结论:MARK2在肝癌组织中高表达,并促进肝癌细胞的迁移。

Abstract: Background and purpose: Hepatocellular carcinoma (HCC) is one of the common malignancies in China. The mechanism of its occurrence is not yet clear. The aim of this study was to investigate the expression of microtubule affinity-regulating kinase 2 (MARK2) in hepatocellular carcinoma and its effect on the migration of hepatoma cells. Methods: The expression of MARK2 in HCC tissues was analyzed by tissue microarray and immunohistochemistry. MARK2 gene eukaryotic expression vector and its empty vector were transfected into hepatoma HepG2 cells, and the expression of MARK2 protein after transfection was detected by Western blot. Transwell assay and cell scratch test were used to detect the effect of MARK2 on the migration ability of HepG2 cells. We analyzed MARK2 expression in liver cancer tissues and normal tissues using the Cancer Genome Atlas (TCGA), and the relationship between MARK2 and clinicopathological features of liver cancer patients. Kaplan-Meier plotter, an online survival analysis software, was used to analyze the relationship between MARK2 expression and prognosis of liver cancer patients. Results: The expression of MARK2 in HCC tissues was significantly higher than that in adjacent tissues. The high expression of MARK2 in HCC tissues was significantly associated with TNM stage and pathological grade (P<0.001). The survival analysis showed that the 5-year survival rate of MARK2 high expression patients was significantly lower than that of MARK2 low expression groups (P<0.01). After the MARK2 gene eukaryotic expression vector was transfected into HepG2 cells, the expression of MARK2 protein was up-regulated, and the migration ability of hepatoma cells was enhanced. Conclusion: MARK2 is highly expressed in HCC and promotes the migration of hepatoma cells.
