胰腺神经内分泌肿瘤中DJ-1蛋白高表达的临床意义

The clinical significance of DJ-1 over expression in pancreatic neuroendocrine neoplasms

摘要:    背景与目的:胰腺神经内分泌肿瘤(pancreatic neuroendocrine neoplasms,PNENs)症状复杂多样,易发生误诊和漏诊。本研究旨在探讨PNENs中DJ-1蛋白表达水平及其在病程进展及预后中的临床意义。方法:采用ELISA法检测16例PNENs及25例健康人血清中DJ-1蛋白的水平,采用免疫组织化学法检测了78例PNENs病理组织中DJ-1蛋白的表达情况。结果:PNENs患者血清中DJ-1蛋白水平显著高于健康人(36.19±6.71vs 24.68±5.94 ng/mL;P<0.001);53.8%的PNENs组织中DJ-1蛋白呈阳性表达(42/78);PNENs组织中DJ-1表达水平与淋巴结转移(P=0.033)、远处转移(P=0.017)、TNM分期(P=0.012)、以及病理分级(P=0.049)呈显著正相关;作为非独立风险因素,DJ-1阳性表达与PNENs患者OS(P=0.003)及DFS(P=0.018)缩短均显著相关。结论:PNENs存在DJ-1蛋白高表达,且与肿瘤的侵袭转移、病程进展及不良预后相关。

Abstract:       Background and purpose: The symptoms of pancreatic neuroendocrine neoplasms (PNENs) are complicated, that might lead to misdiagnosis. This study aimed to detect the expression level of DJ-1 in PNENs and explore the clinical significance of DJ-1 in PNENs. Methods: DJ-1 protien levels in the serum of 16 cases of PNENs patients and 25 cases of healthy persons were detected by ELISA analysis; The expressions of DJ-1 in 78 cases of PNENs tissues were detected by immunohistochemical staining. Results: The DJ-1 level in the serum of PNENs patients was significantly higher than healthy persons (36.19±6.71 vs 24.68±5.94 ng/mL; P<0.001); 53.8% (42/78) cases of PNENs tissues showed DJ-1 positive staining; DJ-1 expression level in PNENs tissues had significantly positive correlation with lymph node metastasis (P=0.033), distant metastasis (P=0.017), TNM stage (P=0.012), and pathology grade (P=0.049); As a dependent risk factor, DJ-1 expression was significantly associated with shorter OS (P=0.003) and DFS (P=0.018) of PNENs patients. Conclusion: High expression of DJ-1 protein correlated with invasion and metastasis, disease progression, and poor prognosis in PNEN.
